چکیده

Thymidylate synthetase from Escherichia coli has been shown to catalyze an exchange between water and the hydrogen on carbon atom 5 of deoxyuridylate at a rate which is approximately 5 to 10% of the rate of the over-all reaction. The exchange reaction exhibited an absolute requirement only for tetrahydrofolate. In the absence of formaldehyde or of Mg++, the exchange activity was reduced to about 25 and 40%, respectively, of the activity of the complete system, whereas the rates of both the over-all reaction and the displacement of tritium from deoxyuridylate-5-3H were reduced to about 5 and 25%, respectively. These findings have been interpreted as support for the reversible formation of an intermediate having a methylene bridge between carbon atom 5 of deoxyuridylate and nitrogen atom 5 of tetrahydrofolate.

A new assay of thymidylate synthetase activity based on the release of tritium from deoxyuridylate-5-3-H.

The tritium atom of deoxyuridylate-5-3H is obligatorily displaced in the thymidylate synthetase reaction and equilibrates with water. Accordingly, a new and simple assay for the enzyme has been devised which is based on the measurement of the tritium released. This method shows essentially the same substrate requirements and pH optimum as the spectrophotometric assay. It has the advantage of mu...

5-Fluoro-2'-deoxyuridylate: covalent complex with thymidylate synthetase.

5-Fluoro-2'-deoxyuridylate causes a rapid inactivation of thymidylate synthetase that is dependent upon prior complexation of the cofactor 5,10-methylenetetrahydrofolate. The enzyme-5-fluoro-2'-deoxyuridylate complex may be isolated on nitrocellulose membranes and is not disrupted by 6 M urea. Upon reaction of 5-fluoro-2'-deoxyuridylate with the enzyme in the presence of 5,10-methylenetetrahydr...

Interaction of thymidylate synthetase with 5-nitro-2'-deoxyuridylate.

5-Nitro-2'-deoxyuridylate (NO2dUMP) is a potent mechanism-based inhibitor of dTMP synthetase. After formation of a reversible enzymeìnhibitor complex, there is a rapid first order loss of enzyme activity which can be protected against by the nucleotide substrate dUMP. From studies of model chemical counterparts and the NO2dUMPdTMP synthetase complex, it has been demonstrated that a covalent bon...

Interaction of deoxyuridylate with thymidylate synthetase.

The reaction of deoxyuridylate with Lactobacillus casei thymidylate synthetase (methylenetetrahydrofolate: deoxyuridylate C-methyltransferase) in the absence of folate or thiols can be followed by changes in the circular dichroic spectra at 267 nm. These changes at the maximum absorption wavelength of the pyrimidine suggest that on binding the interaction of the pyrimidine ring and the deoxyrib...

Nucleotide interconversions. IV. Activities of deoxycytidylate deaminase and thymidylate synthetase in normal rat liver and hepatomas.

Normal and neoplastic rat liver was analyzed for deoxycytidylate deaminase and thymidylate synthetase. For these assays, highly sensitive radiochemical methods capable of detecting the formation of less than 0.001 /~mole of deoxyuridylate and thymidylate were employed. Deoxycytidylate deaminase was found in normal adult rat liver, rat embryo liver, regenerating liver, Novikoff hepatoma, Dunning...
